This product is Evodia rutaecarpa (Juss.) Benth. of Rutaceae, Evodia rutaecarpa (Juss.) Benth. var. officinalis (Dode) Huang or Evodia rutaecarpa (Juss.) Benth. var. bodinieri (Dode) ) Huang's dry near-ripe fruit. From August to November, when the fruit has not yet cracked, cut off the fruit branches, sun-dried or low-temperature drying, and remove the branches, leaves, fruit stems and other impurities.

This product is spherical or slightly pentagonal oblate, with a diameter of 2 to 5 mm. The surface is dark yellow-green to brown, rough, with many punctate protrusions or concave oil spots. There is a five-pointed star-shaped fissure at the top, and a fruit stalk with yellow hairs remains at the base. Hard and brittle, with 5 chambers in the ovary, each chamber has 1 pale yellow seed. The gas is fragrant, and the taste is spicy and bitter.

Evodia: remove impurities.

Preparation of Evodia: mash licorice, add appropriate amount of water, decoction, remove slag, add in net Evodia, after suffocating and draining, fry until slightly dry, take out, and dry in the sun. For every 100kg of Evodia, use 6kg of licorice.
Identification:

(1) This product is brown powder. Non-glandular hairs 2-6 cells, 140-350 μm long, with obvious wall warts, and some cells contain brownish-yellow to brownish-red substances. Glandular hair head 7-14 cells, oval, often with yellowish-brown inclusions; stalk 2-5 cells. There are many calcium oxalate cluster crystals, with a diameter of 10 to 25 μm; there are occasional cubic crystals. Stone cells are round or oblong, 35-70 μm in diameter, and have large cavities. Oil compartment debris is sometimes visible, pale yellow.

(2) Take 0.5g of this product powder, add 10ml of hydrochloric acid solution (1→100), shake vigorously for several minutes, and filter. Take 2ml of filtrate, add 1 drop of potassium mercuric iodide test solution, after shaking, a yellow-white precipitate will be formed; take another 1ml of filtrate, slowly add 2ml of p-dimethylaminobenzaldehyde test solution, heat it on a water bath, and the two liquids are junction. A reddish-brown ring is formed.
Assay

Determine according to high performance liquid chromatography (Appendix VI D).

Chromatographic conditions and system suitability test Use octadecylsilane bonded silica gel as filler; acetonitrile-water-tetrahydrofuran-acetic acid (51:48:1:0.1) as mobile phase; detection wavelength is 225nm. The number of theoretical plates should not be less than 7000 according to the peak of evodiam.

Preparation of reference solution Precisely weigh the appropriate amount of evodiamine and evodiamine reference substance, and add methanol to prepare solutions containing 0.1 mg per 1 ml.

Preparation of the test solution: Take about 0.13 g of this product powder (passed through a No. 3 sieve), accurately weigh it, add 80 ml of methanol, heat under reflux for 50 minutes, let cool, filter, and recover the appropriate amount of methanol from the filtrate and transfer it to a 10 ml measuring bottle , add methanol to the mark, shake well, and that's it.

Determination method Precisely draw 5 μl of the above-mentioned two reference solutions and the test solution, inject them into a liquid chromatograph, and measure.

The total amount of evodiamine (C19H17N3O) and evodiamine (C18H13N3O) contained in this product shall not be less than 0.20%.
